Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7, also known as gp40 and Leu9. CD7 is a 40 kDa receptor belonging to the immunoglobulin gene superfamily. The N-terminal amino acid sequence (aa1–107) is homologous to Ig kappa light chain, while the C-terminal region is proline-rich, forming a stalk that projects the Ig domain.

CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, natural killer (NK) cells, a small subset of normal B cells, and malignant B cells. Cross-linking CD7 modulates T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 associates with phosphoinositol 3′-kinase and its ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic subpopulation of human CD34(+) CD38(-) cord blood cells expressing high CD7 levels has lymphopoietic capacity, generating B cells, NK cells, and dendritic cells, but not myeloid or erythroid cells. CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported

This antibody is reported for use in flow cytometric analysis.

Applications Tested

Tested by flow cytometric analysis of normal human peripheral blood cells. Use ≤5 µL (0.8 µg) per test in 100 µL final volume. Optimal antibody titration is recommended.
Cell number per test: 10⁵–10⁸ cells.

Important Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures engineered for optimized fluorescence and minimal spillover/spread.

Target Information

CD7 (gp40, Leu9) is a 40 kDa immunoglobulin superfamily member expressed on immature and mature T-lymphocytes, T cell leukemias, NK cells, and some B cells. Cross-linking CD7 enhances T and NK cell activation, calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 is a key marker in leukemia diagnostics.
