Product Specific Information

The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor and member of the immunoglobulin gene superfamily.
CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, and a subset of B cells. Cross-linking CD7 modulates T cell and NK cell activity by influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 directly associates with phosphoinositol 3′-kinase, and its ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic subpopulation of human CD34(+) CD38(-) cord blood cells expressing CD7 shows lymphopoietic potential, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells. CD7(+) phenotype marks primitive lymphoid progenitors distinct from pluripotent stem cells.
CD7 also cooperates with CD28 in Treg function; deficiency in both reduces Treg numbers and suppressive activity.

Applications Tested: Pre-titrated for use at 5 µL (0.25 µg)/test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures for engineered fluorescence signatures that minimize spectral spillover.
