Target Information

Signal-mediated nuclear import and export proceed through the nuclear pore complex (NPC), which is composed of approximately 50 unique proteins collectively known as nucleoporins. The 98 kDa nucleoporin (NUP98) is generated from a 186 kDa precursor protein through proteolytic cleavage, producing both 98 kDa and 96 kDa nucleoporins localized to the nucleoplasmic side of the NPC.

Rat studies indicate that the 98 kDa nucleoporin functions as one of several docking site nucleoporins for transport substrates. In humans, the NUP98 gene can fuse with multiple genes following chromosomal translocations in acute myelogenous leukemia (AML) and T-cell acute lymphocytic leukemia (T-ALL). It is located in the imprinted gene domain of 11p15.5, a critical tumor-suppressor region associated with several cancers and syndromes, including Beckwith-Wiedemann syndrome, Wilms tumor, rhabdomyosarcoma, adrenocortical carcinoma, and various solid tumors. Alternative splicing yields several transcript variants, not all of which have been fully characterized.
