Product Specific Information

The monoclonal antibody GAP.A3 reacts with the human major histocompatibility complex (MHC) class I molecule HLA-A3, a member of the HLA-A family.
HLA-A3 has two subtypes (HLA-A3.1, HLA-A3.2) differing at amino acid positions 152 and 156.
MHC class I molecules are expressed on all nucleated cells and associate with β2 microglobulin. They present intracellular peptides to cytotoxic T lymphocytes.

HLA-A3 is expressed by 15–25% of cancer patients and can bind the tumor-associated antigen hTERT (human telomerase reverse transcriptase).

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test).

Target Information

HLA-A belongs to the HLA class I heavy chain paralogues, forming a heterodimer with β2-microglobulin.
These molecules present peptides from the endoplasmic reticulum lumen and are expressed in nearly all nucleated cells.
The heavy chain (~45 kDa) gene contains 8 exons encoding leader peptide, α1/α2 peptide-binding domains, α3 domain, transmembrane region, and cytoplasmic tail.

Polymorphisms within exons 2 and 3 determine peptide-binding specificity, important for bone marrow and kidney transplantation typing.
Hundreds of HLA-A alleles are known.

HLA and MHC antibodies are key tools in Immunopeptidomics, enabling identification and characterization of neoantigens via LC–MS/MS.
