This HMN4-14 monoclonal antibody reacts with mouse Notch4, a member of the Notch receptor family.
Notch receptors are 300-kDa single-pass transmembrane proteins with extracellular EGF-like repeats for ligand binding and intracellular domains involved in signaling.

Upon ligand binding (Delta or Jagged), Notch undergoes sequential proteolytic cleavages by ADAM-family metalloproteases and gamma-secretase. The released Notch intracellular domain (NICD) translocates to the nucleus, forming a ternary complex with CSL and Mastermind, activating transcription.

Notch4 is expressed in CD8⁺ splenic dendritic cells, endothelial cells, and macrophages. A truncated, constitutively active form (Int3) results from MMTV insertion and is linked to mammary gland development and tumorigenesis. Notch4 also contributes to myeloid proliferation, hematopoiesis, and embryonic endothelial development.

The HMN4-14 antibody is reported to crossreact with rat and exhibit activating properties.

Applications Tested: Tested on mouse splenocytes and Notch4-transfected cells at ≤1 µg per test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Titrate for optimal performance.
