Product Specific Information

The RM134L monoclonal antibody reacts with mouse CD252 (OX-40 Ligand), a member of the TNF superfamily. OX-40L is induced on mouse splenic B cells stimulated with anti-IgM and anti-CD40, but not expressed on resting or activated mouse T cells.
Interaction between OX-40 (CD134) and OX-40L is involved in T cell-dependent humoral responses. RM134L inhibits the costimulatory activity of mouse OX-40L transfected cells for anti-CD3-stimulated T cell proliferation.

Applications Tested: Flow cytometric analysis of stimulated mouse splenocytes (≤0.5 µg/test, 100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.

Target Information

TNFSF4 (OX40 Ligand) is a type II membrane-bound, non-covalently linked homotrimeric protein expressed on antigen-presenting cells (dendritic cells, activated B cells) and other cells such as vascular endothelial, mast, and NK cells.
It signals through TNFRSF4, expressed mainly on CD4⁺ T cells and some activated CD8⁺ T cells. TNFRSF4/TNFSF4 interaction provides costimulatory signals essential for effective T-cell activation, enhancing proliferation, survival, and cytokine expression (RANTES, IL-2, IL-3, IFNγ).
This signaling plays a critical role in immunotolerance.
