This antibody detects endogenous levels of CD45 only when phosphorylated at Tyr1218.
Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) ubiquitously expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes. It is absent on non-hematopoietic cell lines and tissues.
CD45 glycoprotein plays a crucial role in lymphocyte development and antigen signaling, regulating Src-family kinases. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
CD45RA is one such isoform with restricted expression among lymphoid subtypes. CD45 isoforms vary in their ability to translocate into glycosphingolipid-enriched membrane domains, depending on cell type and physiological state.
CD45 is essential for T- and B-cell receptor signaling, suppresses JAK kinases to regulate cytokine receptor signaling, and promotes cell survival via integrin-mediated pathways. It also modulates apoptosis and various immunological functions.
