Target Information
The retinal pigment epithelium (RPE) and choroid form a specialized system essential for retinal health and function. Best vitelliform macular dystrophy (Best disease) is an autosomal dominant condition characterized by accumulation of lipofuscin-like material within and beneath the RPE, leading to progressive central vision loss.
This disease is linked to mutations in the Bestrophin gene, encoding a protein with four transmembrane domains localized to the basolateral plasma membrane of RPE cells. Human Bestrophin forms oligomeric calcium-sensitive chloride channels. Missense mutations can reduce or abolish Bestrophin-mediated membrane currents. The human Bestrophin protein consists of 585 amino acids.
