• Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
• After retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
• Suggested positive controls: placenta, lung, or ovarian carcinoma.

Target Information

Nitric oxide (NO) is an inorganic, gaseous free radical that functions as a signaling molecule in various cellular processes such as vasorelaxation, neurotransmission, and cytotoxicity.
NO production is catalyzed by nitric oxide synthase (NOS) enzymes, which oxidize L-arginine to produce L-citrulline and NO.
Three NOS isoforms have been identified:

• nNOS (type I): neuronal NOS
• iNOS (type II): inducible NOS
• eNOS (type III): endothelial NOS

All NOS isoforms contain calmodulin, NADPH, FAD, and FMN binding domains.
