Description:
This 4D10.1 monoclonal antibody recognizes human Syk, a 72-kDa member of the Syk/ZAP-70 family of non-receptor protein tyrosine kinases. Syk is expressed most highly in B cells, with lower expression in immature thymocytes, mast cells, and platelets. It can also be detected in fibroblasts, epithelial cells, hepatocytes, endothelial cells, and neuronal cells. Syk is a major component of signaling cascades downstream of B and T cell antigen receptors and plays an essential role in lymphocyte development. Upon recruitment to immunoreceptor tyrosine-based activation motifs (ITAMs) on antigen receptors, Syk is activated by phosphorylation on multiple tyrosines and phosphorylates proteins such as phospholipase C gamma and BLNK/SLP-65. Abnormal Syk expression has been linked to tumor cell migration and invasion in several cancers.

Applications Reported:
This 4D10.1 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
This antibody has been pre-titrated and tested by intracellular staining using the Foxp3 buffer system and flow cytometric analysis of normal peripheral blood cells. Use 5 µL (0.125 µg) per test (100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test.

Excitation/Emission:
633–647 nm (Excitation), 660 nm (Emission), Laser: Red Laser.

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases that are potential drug targets for allergic disorders and autoimmune diseases. SYK may also play a role in cancer. Syk translocates to the plasma membrane upon B cell antigen receptor (BCR) or the high-affinity IgE receptor (FcεRI) triggering and phosphorylates downstream adaptor proteins, initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, or gene transcription. Syk binds receptor assemblies via its SH2 domains interacting with phosphorylated ITAM motifs, and activation is further enhanced by Src-family kinase phosphorylation.
