• For immunohistochemistry (PFA-fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases.
It contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 shows a cell cycle-dependent localization: perinucleolar expression in G1, nuclear matrix localization in G2, and chromosomal association during M phase.
It serves as a proliferation marker since it is absent in G0 cells but increases steadily from G1 through mitosis.
Ki-67 antibodies are widely used to determine the proliferative fraction in neoplasms, with strong correlation between low Ki-67 index and low-grade tumors.
Commonly used as a neuronal marker for cell cycling and proliferation.
