• Immunohistochemistry (PFA-fixed): incubate antibody for 30 min at room temperature.
• For formalin-fixed tissues: heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and in various normal and malignant cell types.
CD146 functions as a calcium-independent cell adhesion molecule and may also participate in signal transduction through Fyn kinase recruitment and subsequent tyrosine phosphorylation of intracellular proteins involved in cytoskeletal organization.
It plays roles in cell adhesion, endothelial cohesion, and vascular integrity. CD146 expression in melanoma cells may facilitate interaction with vascular components, promoting tumor metastasis. Additionally, CD146 may function as an adhesion molecule during neural crest development and as a receptor triggering phosphorylation of FYN and PTK2, increasing intracellular calcium levels.
