• For Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and other normal or malignant cell types.
CD146 functions as a calcium-independent cell adhesion molecule and may act as a signal transduction molecule through Fyn kinase recruitment, leading to tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
It plays a key role in cell adhesion and endothelial monolayer cohesion in vascular tissues. CD146 expression may facilitate melanoma cell interaction with vascular elements, enhancing tumor spread. It may also act as an adhesion molecule in neural crest cells during embryonic development and as a surface receptor triggering phosphorylation of FYN and PTK2, and increasing intracellular calcium concentration.
