Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in endothelial cells and in other normal and malignant cell types. Beyond its role as a calcium-independent cell adhesion molecule, CD146 acts in signal transduction through recruitment of Fyn kinase and subsequent phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
CD146 contributes to endothelial cohesion and may facilitate melanoma cell interactions with the vascular system, promoting tumor spread. It may also function as an adhesion molecule in neural crest cells during embryonic development and as a surface receptor triggering phosphorylation of FYN and PTK2, leading to transient intracellular calcium increases.
