Target Information
CD146 (MCAM) is an integral membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed at cell-cell junctions in all endothelial cells and in various normal and malignant cell types. CD146 functions as a calcium-independent cell adhesion molecule and may also act as a signal transduction molecule by recruiting Fyn kinase, leading to tyrosine phosphorylation of proteins involved in actin cytoskeleton assembly.
CD146 plays a key role in:
- Cell adhesion and endothelial monolayer cohesion
- Interaction of melanoma cells with vascular elements, promoting tumor spread
- Neural crest cell adhesion during embryonic development
- Surface receptor activity triggering phosphorylation of FYN and PTK2 and transient intracellular calcium increase
