Product Specific Information

  • Reconstitute with 1 mL of distilled water.
  • After reconstitution, store product undiluted at 4°C in the dark.
  • For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
  • Mouse anti-Human HLA-A2 antibody, clone BB7.2 recognizes the human HLA-A2 histocompatibility antigen.

Target Information

HLA-A belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is membrane-anchored and plays a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. It is expressed in nearly all cells.

The heavy chain (~45 kDa) gene contains 8 exons:

  1. Exon 1 encodes the leader peptide
  2. Exons 2 and 3 encode the α1 and α2 domains (peptide-binding)
  3. Exon 4 encodes the α3 domain
  4. Exon 5 encodes the transmembrane region
  5. Exons 6 and 7 encode the cytoplasmic tail

Polymorphisms within exons 2 and 3 determine peptide-binding specificity. Typing for these polymorphisms is routinely used in bone marrow and kidney transplantation. Hundreds of HLA-A alleles have been described.

HLA and MHC antibodies are essential in Immunopeptidomics, aiding in the identification and characterization of neoantigens via HPLC coupled with tandem mass spectrometry.
