Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of the T and B lineage as well as by activated mature T and B lymphocytes. By itself, CD25 binds IL-2 only with low affinity, but associates with CD122 (IL-2Rβ) and CD132 (common γ chain) to form the high-affinity IL-2 receptor. Binding of IL-2 to both receptor classes is inhibited by the PC61.5 antibody. CD25 plays a role in lymphocyte differentiation and activation/proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

eFluor™ 506 is excited by a 405 nm violet laser and emits at 506 nm. Use a 510/20 band pass filter or equivalent. Ensure instrument compatibility.

Target Information

CD25 (IL2 receptor alpha chain, IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The receptor complex consists of IL2RA, IL2RB, and IL2RG chains forming the high-affinity IL-2 receptor. Homodimeric IL2RA or IL2RB chains form low- and medium-affinity receptors, respectively. IL2 expression in mature thymocytes is monoallelic, providing precise regulation. IL2 acts as a growth and differentiation factor and regulates apoptosis. It promotes B cell proliferation, enhances NK cell activity, and inhibits granulocyte macrophage colony formation. Disruption of this gene in mice leads to ulcerative colitis-like disease, indicating its role in immune response regulation. Mutations are associated with IL-2 receptor alpha deficiency.
