Product Specific Information

The 5H4 antibody reacts with mouse CD122, the 90–110 kDa interleukin-2 receptor beta chain. CD122 is expressed by NK cells and some T cells in the periphery and is upregulated by activation. CD122 associates with CD25 and CD132 (common gamma chain) to form the high-affinity IL-2 receptor.
Binding of IL-2 to CD122 is not inhibited by the 5H4 antibody.

Applications Reported:
The 5H4 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
The 5H4 antibody has been tested by flow cytometric analysis of resting and activated mouse splenocytes. Recommended usage is ≤0.25 µg per test (defined as the amount of antibody that stains a cell sample in 100 µL final volume). Cell number may vary from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal assay performance.

Target Information

CD122 (Interleukin 2 receptor) is involved in T cell-mediated immune responses, primarily expressed in the hematopoietic system and exists in three forms:

• Low affinity form: α subunit monomer, not involved in signal transduction
• Intermediate affinity form: α/β heterodimer
• High affinity form: α/β/γ heterotrimer

Both intermediate and high affinity forms participate in receptor-mediated endocytosis and mitogenic signal transduction.
The CD122 gene encodes the beta subunit, a type I membrane protein.
Dysfunction of CD122 is associated with oligoarticular juvenile idiopathic arthritis and rheumatoid factor negative juvenile idiopathic arthritis.
