Recombinant rabbit Superclonal™ antibodies are unique offerings from Thermo Fisher Scientific. They consist of multiple different recombinant monoclonal antibodies, combining the sensitivity of polyclonal antibodies with the specificity of monoclonal antibodies, while maintaining lot-to-lot consistency. These antibodies recognize multiple epitope sites on the target, providing higher detection sensitivity for low-abundance targets. Each lot is produced with a known mixture of light and heavy chains, eliminating biological variability typical of polyclonal production.

Target Information

The KRAB (Kruppel-associated box) domain (~45 amino acids) is a transcriptional repression domain found in numerous transcription factors. Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes exist in the human genome. These proteins repress transcription through interaction with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing a RING finger, B boxes, and a PHD finger.

KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency. It interacts with HP1 (heterochromatin protein 1) and KRAZ1 (Kruppel-associated box-containing zinc finger protein 1), targeting KRAZ1 to centromeric heterochromatin regions containing HP1α, thereby regulating transcriptional repression. Mutations in KAP-1 that bind KRAB but not HP1 lead to random KRAZ1 distribution and strong transcriptional activation.
