The 3F9.D12 antibody reacts with mouse BTLA (B and T lymphocyte attenuator) from both BALB/c and C57Bl/6 strains. BTLA is expressed by peripheral lymphocytes, splenic macrophages, developing B cells in the bone marrow, developing T cells in thymus, and mature dendritic cells (but not immature bone marrow-derived dendritic cells). BTLA acts as a negative regulator of activation and/or function of various hematopoietic cell types. It is reported that BTLA binds to B7-H4, though further studies are needed to confirm this interaction.

Note: The anti-mouse BTLA monoclonal antibody 6F7 stains CD4+ and CD8+ single-positive thymocytes (Hurchla et al). However, other anti-mouse BTLA clones generated simultaneously with 6F7 (8F4, 3F9.D12, 6G3, and 6H6) do not stain SP thymocytes. Despite this discrepancy, clones 8F4, 3F9.D12, 6G3, and 6H6 stain similar populations to 6F7 in splenocytes and bone marrow cells.

Applications Reported: This 6G3 antibody is reported for use in flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg per test (in 100 µL final volume). Cell number should be determined empirically (range: 10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information

BTLA (B and T-lymphocyte attenuator) is a co-inhibitory receptor of the CD28 superfamily. It functions as a lymphocyte inhibitory receptor, downregulating immune responses. BTLA is constitutively expressed on most CD4+ and CD8+ T cells, with expression decreasing upon T cell activation. It remains expressed on Th1 but not Th2 cells. BTLA interacts with the TNFR superfamily member HVEM (herpesvirus entry mediator), forming an important pathway regulating lymphocyte activation and immune homeostasis.
