Product Specific Information
The H74 monoclonal antibody was generated and reacts with human B7-S1, also known as B7-H4 and B7x. Cross-reactivity to other proteins has not been determined. B7S1 is a newly discovered member of the B7 family and may influence T-cell costimulation depending on B-cell activation status. Although BTLA is reported as a counter receptor, further studies are needed to confirm the B7-H4 ligand.
H74 stains human B7-H4 transfected cells but not peripheral blood cells. The exact expression pattern of B7-H4 remains incompletely characterized.
Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of transient hB7-H4 transfected cells. Recommended ≤0.25 µg per test (100 µL final volume). Optimal titration is advised for best assay performance.

Target Information
B7-H4 (also known as B7x, VTCN1) is a costimulatory protein functioning as a negative regulator of T-cell mediated immunity. Although its receptor is unknown, B7-H4 inhibits T-cell proliferation, cell cycle progression, and IL-2 production. It is expressed on activated T-cells, B-cells, monocytes, and dendritic cells, and aberrant expression is linked to human cancers such as lung, breast, and ovarian cancer.
Overexpression of B7-H4 may help tumor cells evade immune surveillance. VTCN1 is also considered a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
