Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting recognition of overlapping epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample when using NovaFluor conjugates in antibody cocktails or single-color controls.

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures that enable engineered fluorescent signatures minimizing spillover and spread impacts.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded by genes on chromosome 11. These subunits associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
