Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. SK7 and UCHT1 antibodies cross-block binding, suggesting overlapping epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.2 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, all part of the immunoglobulin superfamily encoded on chromosome 11. CD3 plays roles in growth arrest, survival, and proliferation of T cells. Expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells. Not expressed on B or NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
