Product Specific Information
The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. Along with the other CD3 subunits (gamma and delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting recognition of overlapping epitopes.
The antibody SK7 recognizes chimpanzee CD3.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.8 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.
Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment with NovaFluor dyes.
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample (regardless of number of NovaFluors). For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures that allow engineered fluorescent signatures with minimized spillover and spread.

Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta chains, which are structurally related immunoglobulin superfamily members encoded on human chromosome 11. CD3 subunits associate with cytoplasmic signal transduction molecules through double tyrosine-based motifs.
CD3 is expressed on:
- 68–82% of normal peripheral blood lymphocytes
- 65–85% of thymocytes
- Purkinje cells in the cerebellum
Not expressed on B or NK cells.
Decreased T lymphocyte percentages may be observed in some autoimmune diseases.
Defects in CD3 genes are associated with CD3 immunodeficiency.
