Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3ε, a 20 kDa subunit of the TCR complex. Along with CD3γ and CD3δ, the ε chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. SK7 and UCHT1 antibodies cross-block binding, indicating overlapping epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for use at 5 µL (0.8 µg)/test on normal human peripheral blood cells (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

The CD3 subunit complex transduces antigen-recognition signals into the cytoplasm of T cells and regulates TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded by closely linked genes on human chromosome 11.

CD3 is expressed on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

Not expressed on B or NK cells. CD3 gene defects are associated with CD3 immunodeficiency and altered T lymphocyte levels in autoimmune diseases.
