Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. SK7 and UCHT1 antibodies recognize overlapping epitopes.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using human peripheral blood cells at 5 µL/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

The CD3 complex transduces antigen-recognition signals and regulates TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits interact with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 is expressed on most T lymphocytes, thymocytes, and Purkinje cells, but not on B or NK cells. Altered CD3 expression may occur in autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
