This antibody detects endogenous levels of p53 only when phosphorylated at Serine 366.
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 upregulates stress-response genes such as p21^waf1^, leading to G1 or G2/M arrest.
p53 consists of four major domains:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at specific residues, including Ser15 and Ser392, is crucial for p53 activation, stabilization, and tumor suppression functions. Elevated p53 levels are linked to stress responses, aging, and cognitive processes such as learning and memory. Mutations in TP53 are common in many human cancers and can lead to loss of tumor suppressor activity. Germline mutations are also associated with cancer-prone syndromes such as Li-Fraumeni syndrome.
