Product Specific Information

This antibody detects endogenous levels of p53 only when phosphorylated at Serine 9.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 upregulation leads to activation of genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at various sites, including Ser15 and Ser392, plays key roles in p53 activation, stabilization, and tumor association. p53 is expressed at low levels in normal cells but elevated in transformed cells, contributing to malignancy. Mutations in TP53 are common in human cancers and can lead to loss of tumor suppressor activity. Germline mutations are also linked to cancer-prone syndromes such as Li-Fraumeni.
