Product Specific Information

Description:
The 24-31 monoclonal antibody reacts with human CD154 (CD40 Ligand, gp39), a 39 kDa transmembrane glycoprotein of the TNF superfamily expressed transiently by activated T cells. CD154 interacts with CD40 on antigen-presenting cells (B cells, monocytes/macrophages, dendritic cells) to mediate T-dependent B-cell activation and cycle entry. Clone 24-31 cross-reacts with cynomolgus monkey CD154.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of stimulated human peripheral blood cells. Recommended use: 5 µL (0.5 µg)/test.

Fluorophore Information:
PE-eFluor® 610 can be excited with laser lines from 488–561 nm and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm / 607 nm / Blue, Green, Yellow-Green Laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD40 Ligand (CD40-L, CD154):
A membrane glycoprotein expressed on activated T cells. Stimulates B cell proliferation and immunoglobulin secretion, induces cytokine production and tumoricidal activity in monocytes, and co-stimulates activated T cell proliferation with IFN-γ, TNF-α, and IL-2 production.
