Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally related to CD28. CTLA-4 is expressed on activated T cells and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28. While CD28 acts as a costimulatory molecule, CTLA-4 inhibits T-cell activation by reducing IL-2 secretion and proliferation. The cytoplasmic domain of CTLA-4 contains ER retention motifs, leading to intracellular localization upon TCR signaling.

This antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.

PE-eFluor 610 is excited by 488–561 nm lasers and emits at 607 nm. Recommended detection filter: 610/20 band pass (PE-Texas Red equivalent). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal loss of brightness and FRET efficiency.

Target Information

CTLA-4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor primarily expressed on CD4⁺ T cells. It binds CD80 and CD86 with higher affinity than CD28 and delivers inhibitory signals to T cells, limiting immune activation. Intracellular CTLA-4 is also found in regulatory T cells and contributes to their suppressive function.
