Product Specific Information
The MR1 monoclonal antibody reacts with mouse CD154 (CD40 Ligand, CD40L), a 39 kDa transmembrane glycoprotein transiently expressed by activated T cells.
CD154 binds CD40 on antigen-presenting cells (APCs) such as B cells, monocytes/macrophages, and dendritic cells, playing a key role in T cell–APC interactions and T-dependent B cell activation.
For flow cytometry staining, stimulate enriched T cells or CD4 cells prior to staining with MR1.
This antibody has been tested by flow cytometric analysis of stimulated mouse splenocytes at ≤0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration is recommended for optimal assay performance.
PE-eFluor 610 can be excited with 488–561 nm lasers and emits at 607 nm. Use a 610/20 band pass filter (PE-Texas Red equivalent).
Protect from light as this tandem dye is sensitive to photo-induced oxidation.
Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Fluorophore performance after fixation may vary; clone-specific validation is recommended.

Target Information
CD40 Ligand (CD154) is a membrane glycoprotein expressed on activated T cells.
It stimulates B cell proliferation and immunoglobulin secretion, induces cytokine production and tumoricidal activity in monocytes, and co-stimulates activated T cell proliferation with IFN-γ, TNF-α, and IL-2 production.
