Product Specific Information

For reconstitution, add 100 µL distilled water to obtain a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step (Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).

Target Information

The KRAB (Kruppel-associated box) domain (~45 amino acids) is a transcriptional repression domain found in numerous transcription factors. More than 220 KRAB-zinc finger protein (KRAB-ZFP) genes exist in the human genome. These proteins repress transcription via interactions with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing a RING finger, B boxes, and a PHD finger.
KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency and interacts directly with HP1 (heterochromatin protein 1) and KRAZ1 (Kruppel-associated box-containing zinc finger protein 1).
KAP-1 targets KRAZ1 to centromeric heterochromatin foci containing HP1α, regulating transcriptional repression. Mutant KAP-1 that binds KRAB but not HP1 leads to random KRAZ1 distribution and strong transcriptional activation.
