Target Information

Delta-like ligand 4 (DLL4), also known as Delta-4, is a homolog of the Drosophila delta protein and functions as a transmembrane ligand for the Notch1 receptor. DLL4 is expressed in the vasculature and plays a critical role in vascular development. It is induced by vascular endothelial growth factor (VEGF) and acts as a negative feedback regulator to control angiogenic sprouting and promote vascular network differentiation.
DLL4 is strongly expressed in tumor vessels of clear-cell renal tumors and bladder cancer. Inhibition of DLL4 increases vascular proliferation but results in defective maturation, leading to decreased tumor growth without apparent toxicity.
