Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-10. Anti-Human IL-10-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect human IL-10 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human IL-10 (500-P20) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant hIL-10.

Western Blot:
To detect hIL-10 by Western Blot analysis, use at a concentration of 0.1–0.2 µg/mL. In conjunction with compatible secondary reagents, the detection limit for Recombinant hIL-10 is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes, with some production by lymphocytes. It plays key roles in immunoregulation and inflammation, including:

• Down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages
• Enhancing B cell survival, proliferation, and antibody production
• Blocking NF-kappa B activity
• Regulating the JAK-STAT signaling pathway

Knockout studies in mice indicate IL-10 is essential for intestinal immune regulation. Mutations in IL10 are associated with increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases related to IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
