Description:
The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. The OKT3 antibody has potent immunosuppressive properties in vivo and has been used effectively in the treatment of renal, heart, and liver allograft rejection. CD3 subunits (gamma, delta, epsilon) are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Crosslinking of TCR initiates intracellular signaling leading to activation and proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.06 µg) per test (100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test.

Target Information

The CD3 subunit complex is crucial in transducing antigen-recognition signals into the cytoplasm of T cells and regulating surface expression of the TCR complex. T cell activation involves CD3 gamma, delta, epsilon, and zeta subunits, which are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with cytoplasmic signal transduction molecules via double tyrosine-based motifs. CD3 may play roles in TCR-induced growth arrest, cell survival, and proliferation.

CD3 antigen is expressed on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

It is not expressed on B or NK cells. Decreased T lymphocyte percentages may occur in autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
