Product Specific Information
The eBioG4.18 monoclonal antibody recognizes the rat CD3 protein, a critical component of the T-cell receptor (TCR) and a marker of the T-cell lineage. CD3 participates in signal transduction from the TCR to the nucleus, inducing responses such as IL-2 transcription and clonal expansion.
- In vitro: Immobilized G4.18 antibody activates T cells; soluble form inhibits allogeneic mixed-lymphocyte proliferation and cytotoxicity.
- In vivo: Demonstrated to induce long-term specific tolerance to organ allografts.
- Applications Reported: Suitable for flow cytometric analysis.
- Applications Tested: Tested with rat splenocytes at ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Recommended careful titration for optimal performance. - Excitation: 633–647 nm
- Emission: 660 nm
- Laser: Red Laser
- Filtration: 0.2 µm post-manufacturing filtered
Target Information
The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits—members of the immunoglobulin superfamily encoded on human chromosome 11.
- CD3 subunits have long cytoplasmic tails associated with signal transduction molecules via double tyrosine-based motifs.
- CD3 plays roles in TCR-induced growth arrest, cell survival, and proliferation.
- Expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes; absent on B and NK cells.
- Decreased T lymphocyte percentages may occur in autoimmune diseases.
- Defects in CD3 genes are linked to CD3 immunodeficiency.
