Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-1alpha. Anti-Human IL-1alpha-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hIL-1alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human IL-1alpha (500-P21A) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant hIL-1alpha.

Western Blot:
To detect hIL-1alpha by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used in conjunction with compatible secondary reagents, the detection limit for Recombinant hIL-1alpha is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is coded by the IL1A gene, signaling through IL-1RI and IL-1RII receptors, shared with IL-1 beta. Located on chromosome 2q13, IL-1 alpha plays an essential role in innate host defense by triggering production of other proinflammatory cytokines and initiating acute-phase responses. Its activity can be moderated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding through competitive inhibition.

IL-1 alpha regulates NF-kappaB and MAP kinase pathways, stimulates IL6 and COX-2 (PTGS2) expression, and enhances nitric oxide (NO) production. Elevated IL-1 alpha levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
