Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-1α. Anti-Human IL-1α-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hIL-1α by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human IL-1α (500-P21A) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant hIL-1α.

Western Blot:
To detect hIL-1α by Western Blot, use at 0.1–0.2 µg/mL. In conjunction with compatible secondary reagents, the detection limit for Recombinant hIL-1α is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through two receptors, IL-1RI and IL-1RII, shared with IL-1 beta.
IL1A is located on chromosome 2q13 and plays a key role in innate host defense by inducing other proinflammatory cytokines and acute-phase responses. IL-1 alpha activity is moderated by IL-1 Receptor Antagonist (IL-1RA), which competitively inhibits receptor binding.
IL-1 alpha regulates NF-kappaB and MAPK pathways, stimulates IL6 and COX-2 (PTGS2) expression, and enhances NO production. Elevated IL-1 alpha levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
