Product Specific Information

The TS2/16 monoclonal antibody reacts with human CD29 (Integrin beta 1), an approximately 130 kDa single-pass transmembrane glycoprotein. CD29 forms heterodimers with various integrin alpha subunits to create the VLA (very late antigen) subfamily of adhesion molecules involved in cell-cell and cell-matrix adhesion. It is broadly expressed on lymphocytes and monocytes, with lower expression on granulocytes. The TS2/16 antibody exhibits activating activity for beta 1 integrins.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and tested on normal human peripheral blood cells
• Recommended use: 5 µL (0.5 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test)

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample before mixing with cells.
• For single-color controls: use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, using novel nucleic acid dye structures engineered for optimized fluorescence signatures with minimal spillover and spread.

Target Information

ITGB1 (Integrin Subunit Beta 1, CD29) is a 110 kDa cell surface glycoprotein expressed by many cell types, including all leukocytes. It forms non-covalent heterodimers with at least six different alpha chains (α1–α6), defining the binding specificity of VLA integrins. ITGB1 plays a critical role in cell adhesion, immune cell trafficking, and regulation of inflammatory responses. It also influences tumor cell drug resistance and cytokine secretion in smooth muscle cells.

There are two major isoforms, beta1A (ubiquitous) and beta1D (expressed in cardiac and skeletal muscle).
