Product Specific Information

The 12G5 monoclonal antibody reacts with human CD184 (CXCR4), also known as Fusin, LESTR, or HUMSTR. CD184 is a G-protein-coupled chemokine receptor that functions as a co-receptor for X4 HIV-1 entry into CD4+ cells. It is predominantly expressed on naive T cell subsets and upregulated by PHA and IL-2 stimulation.

The 12G5 antibody partially inhibits SDF-1–induced chemotaxis and calcium influx, blocks CD4-independent HIV-2 infection, and inhibits CD4-dependent infection by certain T-tropic HIV-1 isolates.

Applications Reported:

Applications Tested:

• Flow cytometry on normal human peripheral blood cells
• Recommended ≤ 0.5 µg/test (in 100 µL final volume)
• Cell count: 10⁵–10⁸ cells/test (empirical optimization recommended)

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It acts as a co-receptor for HIV-1 and HIV-2 entry into CD4+ cells via gp120 binding, mediates CXCL12/SDF-1 signaling, and regulates intracellular calcium and MAPK activation.

CXCR4 is involved in:

• Hematopoiesis and vascularization
• Cardiac and CNS development
• LPS-induced inflammatory response (e.g., TNF secretion by monocytes)

Antibodies targeting CXCR4 can block HIV-1 and HIV-2 fusion and infection of human cells. The amino-terminal domain and second extracellular loop of CXCR4 serve as HIV binding sites.
