Product Specific Information

The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. PD-1 is expressed mainly on activated T and B lymphocytes.

Two novel B7 Family members have been identified as PD-1 ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC). These ligands show overlapping functions and are constitutively expressed on some normal tissues and upregulated on activated antigen-presenting cells.

J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo, both intact and Fab forms of J43 enhance contact hypersensitivity and exacerbate acute GVHD similar to PD-1-deficient cells. J43 injection also exacerbates EAE and NOD diabetes, similar to antibodies targeting PD-L1 and PD-L2.

Applications Reported: Flow cytometric analysis and in vitro functional assays.
Applications Tested: Tested by flow cytometric analysis of mouse Con-A activated spleen cells and PD-1 transfected cells. Recommended ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).
Endotoxin Level: <0.001 ng/µg antibody (LAL assay).

Target Information

T-cell activation is regulated by PD-1 (PDCD-1), an inhibitory receptor expressed on activated T cells. PD-1 binds to ligands PD-L1 and PD-L2, transmitting inhibitory signals that limit immune activation in the absence of danger signals (e.g., LPS). PD-1 deficiency leads to hyperactivated immune responses and autoimmune diseases. PD-1 often migrates at higher molecular weight than predicted in SDS-PAGE.
