Product Specific Information

The 2B11 monoclonal antibody reacts with mouse and human CXCR4 (CD184), also known as Fusin, LESTR, or HUMSTR. CXCR4 is a G-protein-coupled chemokine receptor functioning as a coreceptor for X4 HIV-1 entry into CD4+ cells and plays a role in lymphocyte migration. Its ligand is SDF1, and it is essential for granulocyte progenitor maintenance and B cell development.

Expression differs between species:

• In mouse thymus: restricted to CD4+CD8+ cells.
• In spleen: predominantly on B lymphocytes.

The 2B11 antibody recognizes the amino terminus of CXCR4 and exhibits functional activity. It causes receptor internalization but does not block SDF1a.

Applications Reported: Flow cytometric analysis and functional assays.
Applications Tested: Flow cytometric analysis of mouse splenocytes at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a G-protein-coupled receptor with seven transmembrane domains. It functions as a co-receptor for HIV-1 and HIV-2 entry via CD4+ cells, mediating Env-dependent viral fusion. CXCR4 binds the chemokine CXCL12/SDF-1, increasing intracellular calcium and activating MAPK1/MAPK3 signaling.

CXCR4 roles include:

• Hematopoiesis and cardiac ventricular septum formation.
• Gastrointestinal tract vascularization.
• Hippocampal neuron survival and cerebellar development.
• Mediating LPS-induced inflammatory responses and TNF secretion.

Antibodies against CXCR4 block HIV-1/HIV-2 fusion and infection. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
