Interleukin-1 beta (IL-1β) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is synthesized as a 31 kDa inactive pro-form that accumulates in the cytosol and is cleaved into the active 17 kDa form upon inflammasome activation. This process involves caspase-1 activation, which cleaves pro-IL-1β into its active form. IL-1β lacks a classical signal peptide and is secreted through an alternate mechanism alongside caspase-1. It signals via IL-1RI and IL-1RII receptors, shared with IL-1α, and its activity is moderated by IL-1 Receptor Antagonist (IL-1RA). IL-1β plays a crucial role in innate immunity by inducing proinflammatory cytokines and acute-phase responses. Elevated IL-1β levels are linked to chronic inflammatory conditions, and neutralizing antibodies may have therapeutic potential.
