Product Specific Information
This TULY56 monoclonal antibody reacts with human CD56, also known as Neural Cell Adhesion Molecule (NCAM). CD56 is a highly glycosylated transmembrane molecule expressed by neurons and plays a role in homotypic adhesion of neural cells. In the hematopoietic system, CD56 is expressed on NK cells and a subset of T cells (NKT cells).
Staining with TULY56 does not block binding of CMSSB, suggesting the two antibodies recognize different epitopes. TULY56 performs better after fixation and permeabilization than CMSSB. The antibody crossreacts with Rhesus macaque.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is designed for cytometers with a 355 nm UV laser. Verify instrument compatibility before use.
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.
Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in dark with minimal impact on brightness and FRET efficiency.

Target Information
CD56 (NCAM, neural cell adhesion molecule) is a transmembrane glycoprotein of the immunoglobulin family involved in cell adhesion and neural development. It is expressed in neurons, NK cells, and subsets of T cells, playing roles in cell migration, axonal growth, and synaptic plasticity. Polysialic modification reduces adhesion. CD56 interacts with extracellular matrix components such as laminin and integrins.
It is found on most neuroectodermal-derived cell lines and tumors (retinoblastoma, medulloblastoma, astrocytoma, neuroblastoma) and serves as a sensitive neuroendocrine marker for tumors such as ovarian granulosa cell tumors. Diseases associated with CD56 dysfunction include rabies and blastic plasmacytoid dendritic cell neoplasms.
