Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except circulating erythrocytes and platelets. It plays a crucial role in lymphocyte activation through its tyrosine phosphatase activity.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and validated for flow cytometry on normal human peripheral blood cells
  • Recommended Use: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm and is designed for cytometers equipped with a 355 nm UV laser. Ensure instrument compatibility before use.

Staining Buffer Recommendation:
When using multiple Brilliant Ultra Violet™ or polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light during storage and staining.

Fixation:
Samples may be fixed in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information

CD45 (LCA, Leukocyte Common Antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising approximately 10% of lymphocyte surface proteins. It is absent from non-hematopoietic cells and tissues. CD45 regulates Src-family kinases, T- and B-cell receptor signaling, and cytokine receptor pathways, influencing lymphocyte activation, survival, and apoptosis. Isoforms such as CD45RA differ by alternative splicing and exhibit cell type–dependent expression.
