Product Specific Information
The 3D12 monoclonal antibody reacts with human CCR7 (CD197), a G-protein-coupled chemokine receptor that binds CCL19 and CCL21.
CCR7 plays a key role in T cell migration, distinguishing naive, central, and effector memory populations, and is also expressed on dendritic cells (DCs).
During DC maturation, CCR7 expression increases and contributes to chemotaxis, endocytosis, and survival. Expression can be downregulated upon ligand binding.
Staining Recommendation:
Increase incubation time to at least 45 minutes at 2–8°C for optimal staining.
Applications Reported:
Flow cytometric analysis.
Applications Tested:
Pre-diluted and tested on normal human peripheral blood cells. Use 5 µL (1.0 µg) per test (100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test.
Fluorochrome Information:
Brilliant Ultra Violet™ 661 (BUV661) emits at 660 nm; use with UV (355 nm) laser-equipped cytometers.
Buffer Recommendations:
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific polymer interactions.
Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (00-8222-49) or 1-Step Fix/Lyse Solution (00-5333-54).

Target Information
CCR7, a G-protein coupled receptor, is induced by Epstein-Barr virus (EBV) and mediates effects on B lymphocytes.
It is expressed in blood, bone marrow, lymph node, and intestine, particularly in lymphoid tissues and activated B/T lymphocytes.
CCR7 controls migration of memory T cells and dendritic cell maturation.
Ligand: CCL19/ECL.
CCR family includes CCR1, CCR2A, CCR3, CCR4, CCR5, and Duffy antigen, which are involved in T cell chemotaxis and phagocyte migration to inflammation sites.
