Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a member of the Gata family of transcription factors. Gata-3 is a T cell-specific transcription factor important for thymic development and Th2 differentiation. Expression during embryonic development occurs in the central nervous system, skin, mammary glands, and kidney. Homozygous knockout of Gata-3 is embryonic lethal, indicating its essential role.

Gata-3 is critical for T cell commitment and survival, mainly expressed on CD4 single positive cells in the thymus. During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, inhibiting Th1 differentiation.

Alternative splice variants (50 and 45 kDa) are recognized by this antibody.
Staining requires the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Cross-reactivity with rhesus monkey has been reported.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using mouse thymocytes and the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523).

PE-eFluor® 610 is excited by 488–561 nm lasers, emits at 607 nm. Use a 610/20 band pass filter (PE-Texas Red® equivalent).
Light-sensitive—protect from exposure.
Fixation: Samples can be stored up to 3 days in dark at 4°C using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

GATA3 binds enhancer elements of T-cell receptor genes (alpha, beta, gamma, delta), regulating their expression. Expressed in T cells but not in B cells or macrophages, GATA3 is essential for T-cell receptor gene activation. The human gene spans 17 kb with six exons, encoding two zinc finger domains highly conserved with GATA1.
