Product Specific Information

Description:
The IP26 monoclonal antibody reacts with the alpha beta chain of human TCR, expressed by the majority of peripheral T cells.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended use: 5 µL (0.25 µg) per test in a final volume of 100 µL.
Cell number: 10⁵–10⁸ cells/test (empirically determined).

Fluorophore Information:
PE-eFluor 610 can be excited with laser lines from 488–561 nm and emits at 607 nm.
Recommended filter: 610/20 band pass (equivalent to PE-Texas Red).
Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in:

• 1-step Fix/Lyse Solution (Product #00-5333-57)

Storage up to 3 days in dark at 4°C with minimal impact on brightness and FRET efficiency.

Target Information

The T cell antigen receptor (TCR) consists of a ligand-specific alpha/beta heterodimer associated with invariant CD3 subunits (γ, δ, ε, ζ).
TCR engagement induces T cell activation, differentiation, and cytokine production.
Most human T cells express TCR α/β, while γ/δ T cells are mainly found in epithelial tissues.
Defects in TCR expression affect immune regulation and can be linked to autoimmune diseases and tumor recognition.
Superantigen engagement of TCRs can cause toxic shock syndrome.
Autoantibodies to TCR Vβ segments have been identified in autoimmune diseases such as RA and SLE.
