Product Specific Information

The MFL3 monoclonal antibody reacts with mouse Fas (CD95) Ligand, a 40 kDa type II transmembrane glycoprotein belonging to the TNF family. FasL is expressed by activated mouse T cells, and interaction with its receptor CD95 induces Fas-mediated apoptosis. Human FasL is known to be cleaved by matrix metalloproteinases (MMPs) to generate a 26 kDa soluble form, while mouse sensitivity to MMPs remains unreported.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Validated for flow cytometry using mouse FasL transfected cells and activated T cells at ≤0.25 µg/test (in 100 µL final volume). Optimal antibody titration is recommended.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD178 (Fas ligand, FasL) is a type-II membrane protein with its N-terminus in the cytoplasm and C-terminal region extracellular. Its receptor, FasR, is a type-I membrane protein of the TNF/NGF receptor family. Interaction between FasL and FasR triggers apoptosis via receptor oligomerization and downstream signaling. CD178 exists as both membrane-bound (45 kDa) and soluble (26 kDa) forms, the latter generated through metalloproteinase cleavage.

CD178 is mainly expressed in activated T-lymphocytes and NK cells, as well as immune-privileged tissues (testis, eye). Expression is also found in thymus, liver, ovary, lung, heart, and kidney. Fas/FasL-mediated apoptosis is critical in antiviral immune responses and immune regulation. Dysregulation of CD178 expression is associated with atherosclerosis, inflammatory diseases, AIDS, hepatitis, and cancer.
