Product Specific Information

The AA4.1 monoclonal antibody reacts with mouse C1qRp, a 130–140 kDa C-type lectin-like type I transmembrane protein. The AA4.1 antigen was originally identified on early stages of B cell development in bone marrow. Adult spleen and fetal liver also have detectable numbers of AA4.1⁺ cells.
In combination with other hematopoietic progenitor markers (Thy-1, Sca-1, c-Kit, CD43, CD24), bone marrow Lymphoid-Committed Progenitors (CLP) can be subdivided based on AA4.1 expression.
Surface IgM (sIgM), CD23, and AA4.1 expression define three nonproliferative immature/transitional B cell subsets:

  • T1 (AA4.1⁺ / CD23⁻ / sIgM^hi)
  • T2 (AA4.1⁺ / CD23⁺ / sIgM^hi)
  • T3 (AA4.1⁺ / CD23⁺ / sIgM^lo)

AA4.1 is also expressed in cytoplasmic vesicles of endothelial cells, megakaryoblasts, and platelets. Monoclonal antibodies AA4.1 and 493 recognize different epitopes of the same molecule.

Applications Tested: Flow cytometric analysis of mouse bone marrow and splenocyte cells.
Recommended usage ≤ 0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize antibody titration for best results.


Target Information

The encoded protein is a cell-surface glycoprotein and type I membrane protein originally identified as a myeloid cell-specific marker. It was initially thought to be a receptor for C1q but is now known to be involved in intercellular adhesion and apoptotic cell clearance.
The cytoplasmic tail interacts with moesin, linking transmembrane proteins to the cytoskeleton and aiding cytoskeletal remodeling.
